Efficacy, Safety and Tolerability of Givinostat in Non-ambulant Patients With Duchenne Muscular Dystrophy

This is a randomised, double-blind, placebo-controlled, multicentre study to evaluate the efficacy, safety, and tolerability of givinostat in non-ambulant male paediatric (aged 9 to <18 years) patients with DMD. 138 patients will be randomised 2:1 to givinostat or placebo and will be treated for 18 months. - Planned screening duration: approximately 4 weeks (±14 days) - Planned treatment duration: 18 months (approximately 72 weeks) - Planned follow-up duration: 4 weeks (±7 days) (for patients not participating in the long-term safety study) - Total duration of study participation: up to 83 weeks (ie, 20-21 months)

Benralizumab Patient Access Programme Study: Retrospective Study in UK Severe Asthma Centres

Retrospective secondary data collection observational chart review study. Data collection and analysis will be performed on a cohort of patients who have previously been enrolled on the BPAP. Approximately 300 patients taking part in the BPAP between April 2018 and November 2019 and who meet the eligibility criteria will be recruited to participate in the study. Data for each participant in the study will be obtained retrospectively with a focus on obtaining data at baseline (12 months prior to first benralizumab dose or at first assessment in the severe asthma center if <12 months), Index date (date of first benralizumab dose) and at least up to 24 months after index date. Primary Objectives - To describe baseline demographic and clinical characteristics of severe eosinophilic asthma patients enrolled in the BPAP - To describe background treatment patterns of severe eosinophilic asthma patients at baseline and after benralizumab initiation Secondary Objectives: - To describe clinical outcomes after initiation of benralizumab therapy in severe eosinophilic asthma patients treated with benralizumab at 12 and 24 months - To describe patients' adherence to benralizumab, persistence and discontinuation rates and reasons for discontinuation at 12 and 24 months

Thumb Osteoarthritis Prognosis for Supported Self-managment

Background Thumb base osteoarthritis (OA) is common in adults 40 years and over. It affects a person's ability to work, be independent and care for others. Sufferers complain of severe pain and difficulty in completing everyday tasks. The main treatment for the condition is advice on how a person can manage their condition (self-management), reduce their pain, strengthen their hands with exercises and practical tips on tackling painful tasks, sometimes providing a hand splint for support. The treatment is provided by occupational and physiotherapists. We know that this treatment helps suffers in the short term (up to three months). But the research was done in people with few other health problems and while self-management support helps provide pain relief for most people, there are some people who don't benefit. The aim of this research is to see how pain and other hand problems change over a period of six months after the start of treatment, to understand people's experience of care, and examine why some people improve, and some do not. Research Plan In patients receiving treatment for thumb base OA at four NHS sites, who are willing to take part, this research will: - 1. Record changes in symptoms and quality of life at three and six months from when treatment began in a postal questionnaire/survey. 2. Discuss the experience of care and people's beliefs about what makes treatment a success by interviewing a small group of patients. 3. Analyse patient characteristics, to see if it is possible to determine how they will respond to treatment. 4. Develop recommendations for improving care.
