A Study on the Safety, Effectiveness and Immune Response of Meningococcal Combined ABCWY Vaccine in Healthy Adolescents and Adults

The purpose of this study is to assess the safety, effectiveness and immune response of the meningococcal combined ABCWY vaccine (GSK4023393A) intended to protect against invasive meningococcal disease (IMD) caused by all 5 meningococcal serogroups. The first time-in-human (FTIH), Phase I part of this study will be conducted in healthy adults in a dose-escalating fashion with 2 formulations of the investigational MenABCWY-2Gen vaccine and will serve as a safety lead-in to the Phase II study. The Phase II part of the study will be conducted in 2 parts: The 'formulation and schedule-finding' part will follow in healthy adolescents and young adults and it is designed to select the vaccine formulation and the schedule to be tested in Phase III. The 'blood sourcing' part will be conducted in healthy adults in order to collect sufficient serum samples for the development of assays to be used in the MenABCWY-2Gen vaccine clinical development program.

Topical Lidocaine Anesthesia for Nasopharyngeal Sampling

Introduction: Nasopharyngeal sampling is widely used in the diagnosis of the Coronavirus Disease 2019 (COVID-19). The aim of this study is to evaluate the effects of topical lidocaine application for nasopharyngeal sampling, on pain perception, the comfort of the patients, and the application difficulty for healthcare staff. Methods: This study is a prospective randomized placebo-controlled study conducted with 100 healthy volunteers (50 participants in Lidocaine group and 50 participants in Placebo group). Two ml of a solution containing 10 mg/ml of lidocaine was applied to each nostril of the participants in the Lidocaine group, and the same dose of 0.9% NaCl to the Placebo group. Investigators performed two repeated sampling one hour apart. The first sampling was performed before intervention, and the second was performed five-minutes after intervention. Investigators compared the changes in pain intensity and discomfort intensity using two numerical rating scales, the frequency of undesirable reactions, and the judgment of the practitioner staff about the sampling procedure between first and second sampling.

Liver transplantation has become the accepted treatment option for advanced liver failure in recent years. In addition to the increase in the number of centers performing transplant and the decrease in complication rates depending on the experience gained, the most common biliary complications did not improve at the desired level. For this reason, studies in this field are frequent today. The structural complexities of HLA antigens have been the subject of investigation in recent years. With the development of DNA and RNA amplification methods; It is increasingly difficult for physicians to keep up with the increasing burden of knowledge regarding the genetic aspects of these antigens. Concrete benefits of this gene complex, which has been investigated in various fields and preserves its mystery, have not been determined in liver transplant patients. However, there are publications showing its connection with the bile epithelium in liver transplantation and its relationship with the disappearing bile duct syndrome. In this study, our aim is to explain whether the bile duct complications, which appear like a continuous wall and can lead to frightening pictures, are related to HLA tissue compatibility.
